It has been a long time since the brothers last saw the sisters because Shredder sent them away to Japan. There are times when the brothers are concerned about their welfare and wonder where they are. 
In New York
At Night
The turtles after Snakeweed who was kidnapping people in New York. While on the run Snakeweed grabs an unsuspecting  delivery man. "Not the pizza guy! Take Donnie!" Mikey yelled at Snakeweed. "Snakeweed's getting away!" Donnie yelled at his younger brother. "Somebody help me!" The pizza guy yelled as Snakeweed runs into an alleyway. The brothers stop by Leo putting his hand up. "We've got him trapped." Leo said to his brothers. "Good. Then let's go weed-whacking!" Raph said while taking out his weapons. "Tree-trimming!" Donnie said while taking out his weapon. "Gardening!" Mikey yelled taking out his weapons while his brothers look at him. "Boom! Ha ha! Oh, yeah. Mikey put the sprinkles on that sundae." Mikey said to them making Raph sigh. "Let's go!" Raph said as he walks over to the alleyway but Leo stopped him. "Hang on, Raph. We're not charging a 12-foot regenerating mutant." Leo said to him. "Then what are we doing?" Raph asks him. "We'll take Snakeweed from above. Tie him up in the clotheslines. Come on." Leo said to him and he starts climbing the building. "That'll take forever, and we have to stop Snakeweed now! I mean, he's kidnapping people." Raph said to him making Leo stop and he looks down at them. "Not just people, people who bring pizza." Mikey said to Raph. "We take him from above. Let's move!" Leo yelled at them. Then the brothers stop on the rooftop that so blew Snakeweed. "On my count. One, two-" Leo said and the brothers look down but Snakeweed was gone. "Where'd he-" Leo said to himself. "And the award for worst leader goes to-" Raph said making Leo cut him off. "How am I the worst leader?" Leo asks him. "If we did this my way, Snakeweed would be toast by now." Raph said to him. "Ugh. That'd be some nasty toast." Mikey said to himself as Donnie moves him away from Raph and Leo. "This way would've worked if you hadn't wasted time second-guessing my orders." Leo said to him. "Don't give bad orders, and I won't second-guess 'em." Raph said to him. "You know what, Raph? If you think you could do better, why don't you lead?" Leo said to him. "First good idea you've had all day." Raph said to him and the two start growling at each other. "We'll compromise. I'll lead." Mikey said to them making Leo and Raph glare at him. "Fine, Raph, the team's yours. I'm out of here." Leo said to him and he walks away from his brothers. "I can't believe he's gone." Mikey said to them making Donnie put his hand on Mikey. "Aw, don't worry, Mikey. Leo, uh, he just needs some space." Donnie said to him making Mikey look at him. "You meant the pizza guy, didn't you?" Donnie asks him. "Yeah." Mikey said to him.
As Leo walked from roof to roof, he was irrationally mad. Leo stops by one of the roofs and he looks down at the streets below. Leo then hears something behind making him turn to see the Foot clan. "Foot clan? You guys just made my night." Leo said to them and the clan attacks him but Leo manages to defeat every single one of them in only a matter of seconds. "Ahh. I feel so much better." Leo said to himself but then a female warrior wearing a mask concealing the bottom half of her face leaps downward. "Not bad." The female warrior said to him. "Uh, thanks?" Leo said to her hesitantly. "You might actually be a challenge." The female warrior said to him, she then takes out a sword, which is totally coated in blinding powder and she flashes it directly into his face, causing him to get blinded, cough, and have bloodshot eyes. The female warrior then knees Leo in the chest then easily pins him to the ground, then she aims her sword at his neck. "Guess not." She said to him and she takes off her mask. "My name's Karai. See you around." Karai said to him and she then runs away without revealing any other source of information. "What was that?" Leo asks himself very confused.
In the shadows, Karai notices Jesse leaning against a wall. "And where were you?" Jesse asks her as Karai walks over to her. "I had an encounter with a blue mask turtle," Karai explained to her as she stood before her. "Really..." Jesse said as she emerged from the. "I'll take care of him." Jesse said to her with a little smile.
