Genome-wide association studies have linked thousands of genomic regions to disease risk, but pinpointing the genes that cause disease has been challenging. In Nature Genetics, Saori Sakaue, Soumya Raychaudhuri, and colleagues describe a statistical method, SCENT, that they developed and used to identify likely causal genetic variants for common and rare diseases. The method models association between enhancer activity and gene expression in single-cell data. The team applied SCENT to nine multimodal datasets and a total of more than 120,000 single cells or nuclei, creating 23 cell-type-specific enhancer-gene maps. The method enables the creation of cell-type-specific enhancer-gene maps, which could help define the function of noncoding variants. #BroadInstitute #Science #ScienceNews #Research #ScientificResearch

I'm all for model organisms, but when it comes to understanding human biology, there's no true substitute for studies in humans. Fortunately, genetics provides an observational way of understanding human biology, and the classic way we've collected data at 23andMe (by self-report) makes participation in research relatively easy. In this paper, Xin and friends show that drugs with genetic support from 23andMe are 2-3x more likely to succeed, and if you can get even more confident in your gene mapping, that can go up to 4-5x. https://lnkd.in/gNWeUADs

CRISPECTOR2.0 offers improved accuracy by integrating a reference-free method for detecting allele-specific activity and quantifying off- and on-target effects. Thereby, it addresses the challenge of off-target effects in genome editing, which vary among individuals due to genetic diversity. The human genome contains numerous single-nucleotide variants (SNVs), which contribute to differences between gene copies and among individuals. SNVs at on-target or off-target sites can either enhance or reduce Cas9 activity by modifying the nuclease recognition sequence. However, existing pipelines, including CRISPECTOR (previously featured in CRISPR Medicine News), typically rely on a single reference genome for CRISPR activity analysis or require prior knowledge of the variants (alleles) in the sample.
