Conclusions: In real-world clinical practice settings, a heterogeneous population of patients who received sequential radium-223-taxane therapy had a low incidence of hematologic events, with a median survival of 1 year from taxane initiation. Preoperative systemic chemotherapy response is an independent predictor for OS in CRPM patients selected for CRS/HIPEC with Mitomycin C (35 mg/m2, 90 min) or Oxaliplatin (368 mg/m2, 30 min). Multimodality treatment is key for an oligometastatic process but needs to be carefully balanced with toxicity including personalising the HIPEC agent chosen for these patients. Future trials should consider this finding which also has implications for patient selection and timing of CRS/HIPEC. https://lnkd.in/eym4rvr8

We conduct an open-label, single-arm, phase I/II study of BC19 CAR T cells in 50 patients with R/R MM (ChiCTR2000033567). The primary endpoint was safety. BC19 CAR T cells are well tolerated with grade 3 or higher cytokine release syndrome in 8% of patients and grade 1 neurotoxic events in 4% of patients, which meet the pre-specified primary endpoint. Secondary endpoints include overall response rate (92%), median progression-free survival (19.7 months), median overall survival (19.7 months) and median duration of response (not reached). Our study demonstrates that bispecific BC19 CAR T cells are feasible, safe and effective in treating patients with R/R MM.

And the news is not always good... FibroGen, Inc. announced topline data from the Phase 3 LELANTOS-2 trial of pamrevlumab for the treatment of ambulatory patients with Duchenne muscular dystrophy on background systemic corticosteroids. The study did not meet the primary endpoint of change in the North Star Ambulatory Assessment total score from baseline to week 52 (placebo-corrected mean difference -0.528 points; 95% CI -2.308 to 1.251; p=0.5553). Secondary endpoints measured by change from baseline at week 52 in 4-stair climb velocity, 10-meter walk/run test, time to stand, time to loss of ambulation, and proportion of patients with greater than 10 seconds in the 10-meter walk/run test were also not met. I'm fortunate to be working for Opthea, a great company with a novel drug candidate for nAMD (neovascular, a.k.a. "wet", Age-related Macular Degeneration). OPT-302, "sozinibercept", is the first promising new candidate treatment for this disease in nearly two decades and augments existing standard of care treatments focused on inhibition of the VEGF-A disease pathway. Strong progress for #DSR in #heartfailure on two fronts. The first two patients in our US MOJAVE study have been successfully treated and we are on track to start randomised phase in Q1 24.
