Take a look at our latest article in PNAS (https://lnkd.in/ej-CXuhW), where we employed advanced atomistic simulations to unveil the entire transport cycle (including previously unknown structures) of a homolog of the cardiac Na /Ca2 exchanger—an essential membrane protein implicated in heart disease. Free energy landscapes derived from our simulations (and validated by previous experiments) uncover the molecular basis of the structure-function relationship in this transporter, offering a methodological pathway to explore that same relationship in any other membrane protein within this class. Our results reveal that functional specificity is determined by the viability of an (occluded) intermediate protein conformation.
