Proper storage of HPLC (High-Performance Liquid Chromatography) columns is crucial to maintain their longevity and performance. Let's delve into some guidelines for storing HPLC columns: 1. Short-Term Storage (Overnight): - If you plan to use the same column the next day, you have a couple of options: - Leave the Mobile Phase in the Column: You can leave the mobile phase used in the last analysis within the column. Alternatively, if the buffer concentration in the mobile phase is high (>50 mM), you can allow the mobile phase to pass through the column at a very low flow rate. Skipping column conditioning may be possible in this case, especially for normal phase separations. - Consider Buffer Concentration: Be cautious if the buffer concentration is very high (>0.5 M) because prolonged contact with high-concentration buffers could affect the lifetime of pump parts (e.g., injector and switching valves) and the column itself (especially if the pH is close to the column's limit). - Seal the Column: Always seal the column with the supplied end plugs to prevent the chromatographic bed from drying out. 2. Long-Term Storage: - Solvent Choice: For long-term storage, follow the manufacturer's recommendations. Standard HPLC columns should be stored at room temperature and never in a freezer (exceptions apply to protein-modified affinity or active enzyme reactor columns). - Buffered Eluents: Avoid long-term storage in buffered eluents. If the column was previously used with buffered mobile phase, flush it with HPLC-grade water followed by 100% acetonitrile. This prevents buffer precipitation within the sorbent bed. - Seal the Column: Completely seal the column to prevent drying of the chromatographic bed. 3. Column Material-Specific Recommendations: - Silica-Based Normal Phase Columns: Store these with heptane or another inert solvent. - Reversed Phase Columns: Best stored in mixtures of organic compounds (e.g., acetonitrile, methanol) and water. Ensure that the water content does not exceed 50%.

One of our analytical chemists is performing separation by centrifugation via sedimentation. The lighter particles remain at the top of the container, while the denser particles descend to its base. At the highest level resides the organic phase. Even the middle portion of the upper level is subjected to filtration. During the extraction and preparation of L-homoserine, impurities can often interfere with the process, making it difficult to achieve the desired level of purity. Therefore, desalting and impurity removal processes are necessary to improve the purity of L-homoserine during its extraction and preparation.
