Lab Spotlight: Dr. Lingyan Shi at UC San Diego Dr. Lingyan Shi’s lab focuses on bioorthogonal chemical bond imaging of complex molecular events during aging and disease progression. This work aids in disease detection, diagnosis, and understanding neurodegenerative diseases, cancer, and aging processes. Using the Compresstome Vibratome Dr. Shi’s lab uses our Compresstome vibratome for precise tissue slicing, producing high-quality sections for imaging and analysis. “My students and postdocs have generated impressive results with this instrument,” notes Dr. Shi. Delighted be be able to share our work on the Thrombopoietin Receptor, an important molecule in regulation of blood stem cells and platelet production. Deregulation of the Thrombopoietin signalling pathway can cause a number of blood-related diseases, including a rare group of blood cancers collectively known as myeloproliferative neoplasms, which currently have poor treatment options. We have solved the #cryoEM structure of the ectodomain of the Thrombopoietin Receptor in complex with cytokine, and performed biochemical analyses. The structure revealed a few interesting features, which will be the subject of further work in the Kershaw Lab at WEHI (Walter and Eliza Hall Institute of Medical Research). A new treatment for #cancer is showing promise for #TB as well! Texas Biomed researcher Eusondia Arnett, PhD, and colleagues tested a combination therapy in cellular models developed over the past decade in Dr. Larry Schlesinger's lab. The models represent human granulomas, which are dense clumps of cells that form around the bacteria that causes TB. Granulomas are notoriously difficult for antibiotics to penetrate and kill the bacteria. With this model, researchers now have a tool to more quickly screen potential drugs and therapies against TB in the lab. #Health | 𝗨𝗻𝗰𝗼𝘃𝗲𝗿𝗶𝗻𝗴 𝗡𝗲𝘄 𝗗𝗿𝘂𝗴 𝗧𝗮𝗿𝗴𝗲𝘁 𝗦𝗶𝘁𝗲𝘀 𝗧𝗵𝗿𝗼𝘂𝗴𝗵 𝗘𝗻𝘇𝘆𝗺𝗲 𝗜𝗻𝘁𝗲𝗿𝗮𝗰𝘁𝗶𝗼𝗻 | The Gervasio Lab from the University of Geneva (UNIGE), led by Prof. Dr. Francesco Luigi Gervasio, has collaborated with Erika Pellegrini and Dr. Matthew Bowler from the European Molecular Biology Laboratory (EMBL) Grenoble. They made a groundbreaking discovery concerning the interaction of enzymes which might have profound implications for cancer and Alzheimer's disease treatment. The research highlighted a previously unknown docking site where two enzymes interact and form a face-to-face dimer. Using high-end technologies like cryo-electron microscopy (cryo-EM) and small-angle X-ray scattering (SAXS), they uncovered this 'dance' between two kinases, providing an alternative drug target site to explore. For almost a decade now, our multidisciplinary community at the Marble Center at the MIT Koch Institute for Integrative Cancer Research has developed an arsenal of miniaturized technologies to advance the field of #RNA therapeutics. As our Director Dr. Sangeeta Bhatia and colleague Dr. James Dahlman at the Emory University School of Medicine write in this intro to a special PNAS issue on RNA delivery systems: "sustained, interdisciplinary efforts are positioned to help RNA therapeutics reach their scientific and clinical potential.” At the Marble Center, we continue to believe that miniaturization is the fastest route to engineering better delivery materials for RNA.
