Check our new Nature Communications paper detailing a novel metal-free photocatalytic cross-electrophile coupling that enables C1 homologation and Csp3-Csp3 coupling. 🔗 https://lnkd.in/eQ2RE-K9 Abstract: In contemporary drug discovery, enhancing the sp3-hybridized character of molecular structures is paramount, necessitating innovative synthetic methods. Herein, we introduce a deoxygenative cross-electrophile coupling technique that pairs easily accessible carboxylic acid-derived redox-active esters with aldehyde sulfonyl hydrazones, employing Eosin Y as an organophotocatalyst under visible light irradiation. This approach serves as a versatile, metal-free C(sp3)−C(sp3) cross-coupling platform. We demonstrate its synthetic value as a safer, broadly applicable C1 homologation of carboxylic acids, offering an alternative to the traditional Arndt-Eistert reaction. Additionally, our method provides direct access to cyclic and acyclic β-arylethylamines using diverse aldehyde-derived sulfonyl hydrazones. Notably, the methodology proves to be compatible with the late-stage functionalization of peptides on solid-phase, streamlining the modification of intricate peptides without the need for exhaustive de-novo synthesis. Molecular glues are small-molecule protein degraders with physical-chemical advantages over proteolysis targeting chimeras (PROTACs), but with very little rational for their design. Thus, methods for the efficient and rapid synthesis of librairies are instrumental to the discovery of new molecular glues. In this report, the groups of Kitamura and Erb disclose the use of SuFEX chemistry to quickly synthesize over 3000 analogues of 5’-amino lenalidomide for direct-to-biology screening, and four were resynthesized and shown to degrade the GSPT1 protein. Here is our latest pre-print focusing on the scaled up enzymatic synthesis of 2'deoxy nucleoside analogues. We explore the reaction scope of nucleoside transglycosylase II and demonstrate the scalability of the process to prepare high value nucleoside building blocks. We also establish a structural basis of the mechanism of transglycosylation which proceeds via covalent catalysis. This was collaborative work with Gideon Grogan and Alex Ascham (York), and Admir Salihovic Andrea Taladriz Sender and Paul Hoskisson Strathclyde Faculty of Science Work was funded by BBSRC The Leverhulme Trust #chemicalbiology #oligonucleotide #biocatalysis
