ChomiX Chemoproteomics platform shines in uncovering protein targets for natural products in Chinese medicine. It ingeniously converts these natural products into versatile chemical probes, faithfully reflecting their bioactivities. Deployed within live cells or diseased tissues, these probes seamlessly capture natural product-binding proteins. Leveraging bioorthogonal coupling reactions, we precisely isolate and enrich these target proteins. With high-resolution mass spectrometry, we achieve precise mapping down to the binding pockets. Here is our latest paper on #formate. After early indications published in 2018, we now consolidate the pro-invasive effect of formate in different ex vivo and in vivo models. Furthermore we found that formate impacts lipid metabolism which can be targeted to prevent the pro-invasive effect by formate. Still many open questions! Exciting findings in our latest research paper! We compared conformational search engines Auto3D, CREST, Balloon, and ETKDG for accurately determining global minima in molecular structures. Using CCS values from ion mobility-mass spectrometry, we validated their effectiveness. Our study highlights the importance of selecting the right conformation generation engines for precise molecular structure predictions, impacting drug design and metabolite structure elucidation. To advance the field, we've introduced the open-source Gas-Phase Conformation Library. Incorporation of sp3 character (such as spirocycles) into molecules has been shown in many cases to improve physiochemical, in vitro, and in vivo ADME (absorption,distribution, metabolism and excretion) properties. Bioisosteric replacement of the phenylsulfonamide moiety with a spirocyclic motif could show the effects of a reduction in aromatic ring count, logP, and removal of a highly polar functional group. As a result, this leads to improving brain penetrance through a reduction in topological polar surface area (TPSA) (https://lnkd.in/ehUMQyVD).
