One of the biggest roadblocks for multiplexed imaging in #Fluorescence #Microscopy is the need to use spectrally distinct fluorescent labels for each target. Researchers introduce a workflow for circumventing this limit via a #DeepLearning-based approach for identifying different cellular structures stained with the same fluorophore, demonstrated for 4-channel imaging with two different fluorophores. #SuperResolution single molecule localization #Microscopy #SMLM is a powerful tool providing resolution in the tens of nanometers, however sample preparation requirements are often more stringent. This is especially true when trying to image in a crowded environment such as the #Nucleus. Preprint research introduces an optimized protocol for three-color imaging of chromatin, with their results providing insight into the relative organization of euchromatin and heterochromatin. #Endocytosis is a fundamental cellular process where a portion of the plasma membrane (PM) and the surrounding fluid are internalized. However, many internalized components, such as cell surface receptors, need to be recycled back to the PM. Researchers have discovered a mechanism for fast recycling of internalized receptors from early endosomes to the PM via a “kiss-and-run” partial fusion process – dubbing it the clathrin-associated fast endosomal recycling pathway (CARP). #LiveCellImaging #TIRF #Microscopy was performed in part using a Nikon Ti2-E inverted microscope.
