We want to update you on an exciting application of HTA-Proteases. Krakatoa users performing #denovosequencing of #antibodies have reported saving significant time and money by reducing the number of proteases required to achieve sufficient breadth and depth of coverage and get confident results. See an example below of using our Krakatoa protease in this application. If you’re interested in learning more, click here https://c-bio.link/z3btzd. In addition to de novo sequencing, the CinderBio team presented a poster at ASMS titled "Blood to Biomarker Quantitation in Under One Hour with Rapid Proteomics using a Hyperthermoacidic Protease”. We report how using quick single-step sample preparation of Krakatoa with high throughput dual trapping column achieves “Rapid Proteomics” with unprecedented turnaround for clinical proteomics (< 1 hour). We found a systematic evaluation of this methodology reproducibly quantified over 160 proteins from just 1 microliter of whole blood. Furthermore, HTA-Protease reaction conditions stabilize Angiotensin I and II peptides permitting detection and quantitation of these bio-active peptides in under half an hour. A pre-print of the work is available here: https://c-bio.link/ggrrs1. Lastly, based on feedback from all of you, we have revised our suggested protocols for using Krakatoa and Vesuvius. Click here to download (https://c-bio.link/wrba4z). Highlights include: Enzyme Dose · Change of suggested Krakatoa dose to a range from 0.2U – 2.0U / ug (note that 1 U = 8 nanograms of enzyme). Lower doses require longer digestion times. 2.0 U / ug was used in the above “Blood to Biomarker” work to achieve digestion times of 20 minutes. · Change of suggested Vesuvius dose to a range from 0.6U – 6.0U / ug (note that 1 U = 8 nanograms of enzyme). This is a substantial change from the previous guidance. Searches · A reminder, alkylation is not required because at pH 3.0 during digestion Cysteines will remain protonated. If you do alkylate anyway, please make carbamidomethylation a variable modification. That’s it.
