Generation and modification, using phage integrases not previously used in S. pombe, a minichromosome with the aim to alter regions of the fission yeast centromere and further elucidate aspects of centromere function. Used Dam-ID to obtain structural information about the fission yeast centromere and binding regions of the centromeric protein Cenp-C, and compared the use of the technique to the more commonly used ChIP.
