  • The role of heparan sulfate proteoglycans in mouse embyronic stem cell differentiation and proliferation

Embryonic stem cells have therapeutic potential in the treatment of various medical conditions, including tissue regeneration and wound repair. Embryonic stem cells have therapeutic potential in the treatment of various medical conditions, including tissue regeneration and wound repair. Current challenges impeding its clinical application include maintenance of pluripotency and directing lineage-specific differentiation. In this regard, the extra-cellular matrix within the stem cell niche has been suggested to play an important role. In particular, the heparan sulfate proteoglycans (HSPGs) have been demonstrated to interact with and potentiate the effects of many classes of growth factors, including the family of FGFs. Mouse embryonic stem cells (D3) are used here as an investigational model. The objective of this study is to analyze the characteristics of HSPGs in differentiated and nondifferentiated mESCs and investigate the role of HSPGs in affecting growth patterns. Real-time PCR was used to quantify relative gene expression and absolute cell numbers were counted using a fluorescent-activated cell sorter (ViaCount assay, GUAVA Technologies). HSPGs from osteogenic differentiated mESCs were also extracted for future studies using high performance liquid chromatography (HPLC). From the proliferation assays, it was discovered that inhibition of HSPG sulfation by 4mM sodium chlorate stimulated growth over control cells. Gene analysis showed that mRNA expression of Syndecan-2 was differentially regulated in osteo- and control differentiated cells, and that enzymes involved in HSPG synthesis, namely Ndst-1,Ndst-3, HS-6Ost1 and HS-6Ost2, were differentially expressed in control- and osteogenic-differentiated cells.
