Yesterday I had the privilege of attending a workshop hosted by Sonja Beken of the European Medicines Agency 3Rs Working Party. The focus was on Qualification of MicroPhysiological Systems, including Organ-on-Chip (OoC) Models, for Specific Contexts of Use to be Applied in the Pharmaceutical Area. I was particularly drawn to the interactive session on cardiotoxicity, where I was impressed by the insights shared by Paola Occhetta, Stefan Braam, and Jean-Pierre Valentin. Their discussions on the lessons learned from CiPA, a collaborative initiative to engineer an assay for assessment of the proarrhythmic potential of new drugs, were enlightening. My main take-home message from those discussions and the other interactions is that the workshop participants (be it OoC developers, pharma users, or regulators), are all part of the same ‘team’. We share a common belief that human complex tissue culture models can significantly impact regulatory science, potentially revolutionizing how we determine the safety of drug candidates for clinical trials. However, it’s crucial that we channel our energy and attention not just inwardly, but also outwardly. We need to lean into our strengths and play our roles effectively. Regulators can push for official commitment to give human complex in vitro models a formal place in IND (Investigational New Drug) Applications. The road towards this commitment can be cautious and lengthy, but the essence is that the use of human in vitro models will need to become a hard requirement in some form. Developers need to ensure that throughout their R&D process their models, protocols, and data align with what regulators will come to expect in terms of robustness, reproducibility, and realistic applicability. A key aspect here is proving applicability to a specific ‘context-of-use’. Pharmaceutical companies can work to proactively provide well-curated, quality-controlled reference compounds to model developers, even if this happens via neutral, non-profit third parties. The use of reference compounds is an essential aspect in proving the applicability of a new model to a particular context-of-use. In conclusion, while it’s hugely beneficial to connect with our ‘team-mates’ at workshops, we must also remember to take on challenges across the board in our own respective realms of influence. Only by acting as a cohesive community can we effectuate the change we believe in. #3Rs #MPS #Organonchip #ooc #organonachip #3RsWP
