Viral infection in fruit flies can alter intestinal homeostasis and lead to physiological changes. Researchers from Institut Pasteur and Institut Curie in Paris employed fluorescently labeled #DNA #oligonucleotides printed by enzymatic DNA synthesis (#EDS) to track viral infection of the fruit fly gut using a technique called RNA FISH. The group found that infection of flies with the Drosophila A virus (DAV) led to changes in intestinal homeostasis, including an over-proliferation of intestinal stem cells (ISCs). The group found that DAV infection caused cytokine release through a JNK signaling pathway, leading to ISC proliferation through growth factor signaling pathways. Sting-Relish NF-κB signaling was found to be required for the ISC proliferation, and the over-proliferation of ISC cells within the fruit fly #gut led to a shortened #lifespan. A marker for age-associated intestinal dysplasia was upregulated in infected flies. #Apoptosis, or programmed cell death, is a mechanism by which the body may eliminate unnecessary, dysfunctional or highly damaged cells. One mechanism of triggering apoptosis involves the clustering of death receptors on the cell surface. A team at the Karolinska Institutet in Stockholm, Sweden recently published a study in which #DNA origami was used as a switchable control to induce apoptosis in #cancer. Reporting in the journal Nature Nanotechnology, the researchers created a DNA origami which contains ligands that, when exposed, bind and cluster death receptors together, triggering cell death. Under the slightly basic pH conditions of normal cells, the ligands are hidden. However in the lower pH conditions of a solid tumor, the ligands are exposed, causing tumor cells to die. The approach was used in mice containing human breast cancer #xenografts and showed up to a 70% reduction in tumor growth.
