🌐 What Are Spheroids? 🌐 Spheroids are transforming how we study cellular behavior. Unlike traditional 2D cultures, these 3D cell aggregates closely resemble the natural architecture and function of tissues. This similarity makes them invaluable for research that requires realistic cell-cell and cell-matrix interactions. 🧫 What Exactly Are Spheroids? These compact, spherical structures are formed when cells are suspended in a culture medium and naturally aggregate. The process is driven by cell-cell interactions, such as integrin-ECM binding and cadherin expression. The result? A microenvironment that can include various cell types—epithelial, mesenchymal, endothelial—mirroring what we find in actual tissues. Spheroids can be created using cell lines, single cells, or even tissue-derived cells from biopsies. This versatility makes them a powerful tool for biomedical research! 🔬 🧬 Why Are They So Special? One key characteristic is their ability to develop distinct layers, similar to the spatial organization in solid tumors or tissues. These layers often include: Outer Proliferating Zone: Active cell growth and division. Intermediate Quiescent Zone: Cells in a state of dormancy. We are proud to say that the project of our scientist Joanna Nowacka will be financed by the directive of the Free State of Thuringia on the Promotion of Research, Technology and Innovation (FTI, Project nb. 2024 VFE 0050). Joanna will be working on a vascularized liver-on-chip model for personalized compound testing (Vascu Chip). Within her work, Joanna will be developing stem cell cultures into “pluripotent” stem cells, from which she can then differentiate multiple cell types. Joanna says: “I believe organ-on-chip technology represents the future of drug testing models. I am eager to join Dynamic42 to contribute my stem cell expertise to their already remarkable platforms.”

Investigating the pharmacokinetic properties of drugs in a biochip-based organ model. This was what we had in mind 18 months ago when we launched a project funded as part of the Sanofi iDEA-iTECH Award Program. Recycling, viability and vascular permeability of two frequently used mAbs were analyzed using our liver-on-chip model. “We were able to verify the recycling process of therapeutic antibodies without compromising cell viability or tissue integrity. Our model helps to overcome the species specificity of animal models and thereby can reduce the frequency of failures in clinical trials.” Anne-Katrin Bothe Senior Scientist at Dynamic42 GmbH, summarizes the results. Initial results were presented at this year's MPS World Summit in Seattle in mid-June and at the annual meeting of the European Organ-on-chip society (EUROoCS) in Milan at the beginning of July. The project partners aim to continue their joint efforts on innovative organ-on-chip technology for the characterization of the disposition of new therapeutic antibodies.
