We are excited to share our latest paper in a collaborative effort with MeltPrep and Celanese Our project showcases the capabilities of the MeltPrep VCM Filament Molder, an innovative tool that produces filaments in extremely small quantities (as little as 3 grams) and works with challenging materials, like low-viscosity EVA28 grades, that are difficult to process with traditional horizontal hot melt extrusion. This development expands the material options for fused filament fabrication, granting access to materials that were previously inaccessible due to limitations in producing high-quality printing filaments. Excited to share our latest publication! Huge congratulations to Christina Glader and co-authors for this amazing work! 🎉 In this research, we tackle a key challenge in drug screening: optimizing the delivery of drugs with poor solubility or permeability, especially when targeting intracellular components. Our study focuses on nanostructured lipid carriers (NLC) and their cellular uptake using high-throughput screening based on acoustic droplet ejection (ADE) technology (Echo liquid handler). Key findings include: 🔹 Surface tension and viscosity of NLC dispersions must be tailored for reliable ADE transfer. 🔹 Automated uptake studies, confirmed by imaging-based and spectral flow cytometry, highlighted the importance of optimizing the matrix composition for cellular uptake. Last week, at the 16th Annual Conference of the European Paediatric Formulation Initiative (EuPFI), Simone Eder delivered an exciting talk titled “𝗣𝗿𝗶𝗻𝘁𝗲𝗱 𝗱𝗿𝘂𝗴-𝗹𝗼𝗮𝗱𝗲𝗱 𝘁𝗲𝗺𝗽𝗼𝗿𝗮𝗿𝘆 𝘁𝗮𝘁𝘁𝗼𝗼𝘀 – 𝗔 𝗻𝗼𝘃𝗲𝗹, 𝗵𝗶𝗴𝗵𝗹𝘆 𝗮𝗰𝗰𝗲𝗽𝘁𝗮𝗯𝗹𝗲 𝗽𝗿𝗲-𝘁𝗿𝗲𝗮𝘁𝗺𝗲𝗻𝘁 𝗼𝗽𝘁𝗶𝗼𝗻 𝗳𝗼𝗿 𝗽𝗮𝗶𝗻-𝗳𝗿𝗲𝗲 𝘃𝗮𝗰𝗰𝗶𝗻𝗮𝘁𝗶𝗼𝗻𝘀 𝗳𝗼𝗿 𝗰𝗵𝗶𝗹𝗱𝗿𝗲𝗻.” This innovative method not only administers medication through the skin but also decorates it with fun, personalized designs. In the pharmaceutical world, lipid excipients play a crucial role, yet their application in 3D-printing, particularly filament-based 3D-printing, remains largely uncharted territory due to their challenging processability. In our latest publication, our scientists showcase the pioneering successful utilization of novel lipid excipient in fabricating advanced oral solid dosage forms via filament-based 3D-printing. Despite the demanding nature of this research, we are genuinely thrilled about the promising prospects it holds for the future of pharmaceutical manufacturing and personalized healthcare. The tableting of biologics is becoming increasingly popular; however, due to the sensitivity of proteins, the formulation and processing of tablets containing biologics have proven to be challenging. Ensuring the stability and efficacy of these sensitive biologics during the manufacturing process is critical. This paper investigates the effects of formulation, tableting, and spray-drying on the stability of catalase. Different saccharides (trehalose, dextran, and mannitol) are evaluated as potential stabilizers. The enzymatic activity, protein aggregation, and structural changes of catalase in non-spray-dried and spray-dried formulations (blends and tablets) are assessed. The study highlights the importance of optimizing both formulation and processing parameters to maintain the stability of biologics in tablets.
